Geeks Give Back: It’s time for the technology community to step up and give!

This is embarrassing to admit. While the Seattle area is first in the nation in percentage of residents who give to charities — over 65 percent donate more than $25 annually — the tech segment remains an elusive donor group within the region.

Non-profit development officers — faced with a divided city of extreme wealth and extreme poverty — routinely ask: “How do we crack the code? How do we get the average tech worker to give?”

And this is why GeekWire — in partnership with Bank of America — launched our Geeks Give Back effort three years ago. We wanted to encourage everyone in the tech industry — from the software developer to the CEO — to think deeply about helping those in need. This year, we are tackling the thorny issues of homelessness and poverty, and we ask you to go here to donate to Geeks Give Back.

Alleviating homelessness, poverty and a lack of access to education in King County are issues we can all agree on. And I believe — subtracting the highest levels of philanthropy from the likes of Bill and Melinda Gates; Connie and Steve Ballmer; and more recently Jeff and MacKenzie Bezos — the tech industry is not pulling adequate weight compared to other industries. As innovators and creators, we should be punching well above our weight.

We need to do more.

A well-researched analysis on homelessness from McKinsey indicated that much of poverty exists because of wealth creation.

Don’t get me wrong. I love tech, but the facts are clear. The robust influx of tech workers to the Seattle region — with great benefits to the overall economy, intellectual capital and culture — has contributed to housing affordability problems. With every $100 increase in average rent, homelessness rises by 15 percent. Cause and effect. No judgment here. It just is.

While the rising tide has lifted many boats, some people don’t even have boats.

So what’s the answer?

Here’s my simple ask: How about giving a little?

There are so many things we say “no” to each day, but we need to tackle these societal problems head-on. And we can’t just keep pushing them under the rug. And so I challenge you to say “yes.” That’s not always an easy path, given the constraints on time, budgets and a wide array of noteworthy organizations.

The Geeks Give Back program — which has raised more than $2 million over the past three years — offers an easy path to do some good, supporting organizations such as Mary’s Place, Mercy Housing, DESC, Plymouth Housing Group and Housing Development Consortium.

And it’s time for our community to step up and do more.

If you are still on the fence, let me offer this. Giving back provides multiple dividends — you feel more connected to your community; you shift perspectives; you pay more attention to issues that matter; and you join an effort to be part of the solution. I’ve personally seen these benefits as campaign chair for the United Way of King County.

As author Sue Monk Kidd notes: “Empathy is the most mysterious transaction that the human soul can have, and it’s accessible to all of us, but we have to give ourselves the opportunity to identify, to plunge ourselves in a story where we see the world from the bottom up or through another’s eyes or heart.”

So, c’mon geeks. On this day of giving and thanks, take the plunge with us. Please, consider GIVING BACK!

Aadhaar not mandatory for any national entrance exam, for now

New Delhi: The Supreme Court on Wednesday said unique identification number Aadhaar cannot be made mandatory for any national entrance examination for now.

The interim order was passed by a bench headed by Chief Justice Dipak Misra in a challenge to making the 12-digit number mandatory for National Eligibility cum Entrance Test (NEET) 2018 for admission to medical courses.

The court said other forms of identification such as voter ID, passport or ration card can be used to register for or appear in the coming NEET. The last date to submit applications for NEET conducted by the Central Board of Secondary Education (CBSE) is 9 March.

This means even a candidate who has Aadhaar is not required to produce it for appearing in national entrance tests like NEET.

Apart from sparing NEET candidates from mandatorily furnishing Aadhaar, the interim order also prohibits CBSE, Indian Institutes of Technology (IITs) and the University Grants Commission (UGC) from making the unique number mandatory for examinations like Joint Entrance Exam (JEE-Main) and National Eligibility Test (NET) and the Common Admission Test held by Indian Institutes of Management (IIMs).

The court also directed the centre to inform CBSE to follow the order and upload it on its website so that students are better informed. Currently, CBSE is the test delivery partner for most national entrance tests, including JEE-Main, NEET, UGC-NET and Central Teacher Eligibility Test.

Earlier this year, CBSE had issued notifications making Aadhaar must for appearing in several national entrance tests.

“The use of Aadhaar for applicants of UGC-NET(July) 2018 will result in accuracy of applicants’ details. This will also help ascertain identities of applicants at the examination centres in a convenient and hassle-free manner,” CBSE had said in one of its notifications dated 23 February, justifying the use of Aadhaar.

However, attorney general K.K. Venugopal told the court that the Unique Identification Authority of India (UIDAI) has not authorized CBSE to make Aadhaar mandatory for students to enrol for NEET.

A Constitution bench comprising Chief Justice Misra and justices D.Y. Chandrachud, A.K. Sikri, A.M. Khanwilkar and Ashok Bhushan is hearing the case against the Constitutional validity of Aadhaar.

The order was passed on an appeal by one Abidali Yusufbhai Patel, who had specifically challenged Aadhaar being made must for filling up NEET enrolment forms. The matter was mentioned in the morning and was taken up for hearing in the afternoon, along with the main challenge to Aadhaar that is being heard by the Constitution bench.

Patel had challenged a 27 February order passed by the Gujarat high court, which refused to grant interim relief to students seeking exemption from submitting Aadhaar while appearing for JEE(Main), 2018 (JEE) and NEET.

During the course of arguments, a challenge to Aadhaar being passed as a money bill was raised by lawyer and former finance minister P. Chidambaram, who carved out the distinction between a money and financial bill. Chidambaram is one of the lawyers representing the petitioners.

“A money bill cannot be introduced in Rajya Sabha. In this case, Rajya Sabha becomes only a recommending house. They have no legislative power but only recommendative power,” Chidambaram told the court.

A batch of petitions have been tagged by the Supreme Court to be heard by the Constitution bench. They challenge several aspects of Aadhaar, the 12-digit unique identity number that has become a bedrock of government welfare programmes, the tax administration network and online financial transactions, and the use/sharing of personal data collected by the UIDAI.

There are more than 1.2 billion Aadhaar holders in the country.

New Delhi: The CBSE has clarified that it has no role in deciding eligibility criteria for medical entrance exam, NEET, and grievances, if any, should be submitted to the Medical Council of India (MCI).

The clarification came following several complaints received by the Central Board of Secondary Education (CBSE) about barring open school candidates and those with biology as an additional subject in Class 12 from appearing for the National Eligibility Entrance Test (NEET).

“The responsibility of CBSE is limited to holding the NEET (UG) examination, based on the eligibility criteria provided by MCI. CBSE has no role to play in deciding the eligibility conditions,” the board said in an advisory.

Students who have pursued schooling through National Institute of Open Learning (NIOS)/ State Open School or those who studied biology or biotechnology as an additional subject in Class 12 are ineligible to appear in NEET.

“Therefore, all the grievances received by CBSE on these issues are disposed of. Candidates are requested to kindly read the information bulletin and FAQs hosted on NEET website before sending grievance to the board in any form,” it added.

This year, NEET will be held on 6 May. Online application process began on 8 February and 9 March is the last date to register. The last date for successful payment of fee online is 10 March till 11.50pm.

Peptides offer fresh insights for cancer diagnosis and treatment

Genes and proteins play essential roles in the maintenance of health and the development of disease and are the focus of the fields of genomics and proteomics, respectively.

Genes, which are composed of 4 nucleic acids, provide the blueprint for constructing all living forms, while proteins, which are composed of some 20 amino acids, are the body’s tireless day laborers, building organs and tissues, forming a complex defense network of antibodies, transporting essential materials to far-flung destinations in the body and facilitating essential chemical reactions.BRCA1 is a human gene that produces tumor suppressor proteins, which repair damaged DNA and help maintain the stability of each cell’s genetic material. Mutations in this gene (and the related gene BRCA2), can disable this repair network, resulting in an increased risk of breast cancer. In the graphic, tumor-associated proteins resulting from mutations in BRCA1 are degraded into smaller fragments or peptides, which circulate in the bloodstream. These circulating peptides are then enriched using specialized nanoporous silica thin films or NanoTraps, then profiled using mass spectrometry to identify among four clinical groups. Less well known are the peptides, though they too are central players in life processes and can shed new light on a variety of diseases. Indeed, the rapidly expanding field of peptide research or peptidomics is poised to deliver fresh biological insights and new methods for the detection and treatment of a broad range of disorders, particularly cancer.

A new book, “The Enzymes: Peptidomics of Cancer-Derived Enzyme Products,” explores the peptidome in keen detail. The particular focus of the book is how peptides, small segments of linked amino acids, can provide researchers and clinicians with vital clues about cancer prognosis currently unavailable through conventional diagnostic methods.

Tony Hu, a researcher in Arizona State University’s Virginia G. Piper Biodesign Center for Personalized Diagnostics, co-edited the new book.

“Back in 2016, I was invited to UCLA to give a seminar. I talked with a professor there — Fuyuhiko Tamanoi — who served for many years as the editor for the book series ‘Enzyme,’” Hu said. “After he heard a bit about emerging peptide research, he decided it would make a fine topic for a new book, so we invited experts worldwide, from Europe, from China, Japan and the U.S. to explore different aspects of the peptidome.”

As Hu explained, the book is divided into three major areas. The first deals with the discovery of biomarkers — peptide signals present in blood or saliva that can be used for diagnostic purposes. The second part deals with peptide biofunction, which turns out to play a critical role in cancer. The third section is devoted to clinical application and regulation of peptide biomarkers. The book therefore spans research on the peptidome reaching from the laboratory bench to the patient bedside, making it the most comprehensive handbook of leading edge peptidomics to date.

Probing the peptidome

Peptides, like proteins, are composed of amino acids. They are generally far smaller molecules, typically composed of just 50 amino acids or less. They are often produced through the breakdown of larger, more complex proteins. This process of protein degradation is a common feature in cancer progression, invasion and metastasis. In the course of these processes, peptide byproducts of the tumor microenvironment circulate in the bloodstream, providing a potentially rich source of biomarkers for disease.

Although there are myriad cancer-specific proteins secreted by active tumors, which could potentially serve as biomarkers, the challenges of monitoring such proteins — particularly in a clinical environment — are often formidable due to fluctuations in their location and abundance. Hu believes that tumor-secreted peptides, which circulate in the blood, can provide an alternate window into underlying activity, providing more fine-grained diagnoses of a given cancer than can be gleaned from either the genes or proteins alone.

 A new field is born

“About 15 years ago, people started looking at peptides,” Hu said.

Peptides are a rich source of information about the proteins they are derived from and also can shed light on the body’s enzymatic network. As Hu explained, enzymatic activity is often a crucial component in cancer progression, but getting a precise handle on tumor-related enzyme activity has, until recently, proved challenging.

“We published a paper in Clinical Chemistry back in 2014,” Hu said. “There’s an enzyme that is pretty active at the local tumor site in breast cancer. When we profile this enzyme on the tumor tissue, it’s showing this dramatically increased expression compared to normal.”

The enzyme, however, dissolves in the blood and is undetectable. Testing such cancer patients shows blood enzyme levels close to normal.

This presents clinicians with two options: 1) Use a very invasive method to biopsy the tumor tissue in order to profile the enzyme directly, or 2) look at the peptide product produced by the enzyme, which can be profiled directly from the blood. When Hu and colleagues used this approach, they found a variation in peptide expression that precisely matched the enzyme activity at the tumor site. The results encouraged Hu that peptides indeed have much to tell us about underlying disease.

In other cases, the peptide itself is malfunctioning, which may be correlated with tumor progression. Further, peptides can provide vital clues concerning a tumor’s surrounding microenvironment, for example signaling a condition of low oxygen or hypoxia, an important precursor of cancer metastasis and an area of research Hu is currently involved in.

Another critical peptide under study is Hepcidin, a 25-amino acid peptide synthesized in the liver, which serves as the principle regulator of iron metabolism in vertebrates. It can also be used as a telltale indicator of elevated iron status, inflammation and infection associated with diseases including cancer and HIV.

In order to profile peptides from blood samples, mass spectroscopy is used, a technique allowing peptides to be accurately identified based on their molecular weights. Once an exotic technology confined to sophisticated laboratories, cutting-edge mass spec technology is now common in hospital settings around the world, further enabling peptide biomarker discovery to advance and enter routine clinical use.

Finding new avenues

As the authors note, specialized enzymes involved in protein degradation — known as proteases — are involved in all phases of cancer progression including early growth, angiogenesis, inflammation, survival and invasion. Their peptide byproducts offer insights into all of these processes. Evaluation of inflammation through peptide analysis could also be used as a rapid and direct indicator of drug toxicity in chemotherapy, a process that currently must rely on patient symptomatology.

Finally, advances in biotechnology have allowed a new class of safe and efficacious peptide drugs to be developed, furthering the aims of personalized medicine. Immunotherapy, in which the patients’ own immune system is used to attack cancer cells, is just one area where peptides are being applied therapeutically. The immune system responds to a cancer-linked peptide by mounting a robust defense — a technique currently being explored in the fight against prostate cancer.

“For these reasons, we believe peptides have been undervalued for a long time in cancer studies,” Hu said.

Accurate, early diagnostic and prognostic markers are still lacking for most cancers, posing the most significant challenge for successful diagnosis and treatment. Vital information, unavailable through proteomic study, is now becoming accessible through sensitive detection of circulating peptides. The new book throws light on an exciting and hopeful domain of research.

With the launch of TESS, NASA will boost its search for exoplanets

NASA is stepping up its search for planets outside our solar system. Its next exoplanet hunting telescope, the Transiting Exoplanet Survey Satellite (TESS), is due to launch from Cape Canaveral on the evening of April 16.

Following the Kepler space telescope’s discovery of more than 5,000 possible exoplanets since 2009, TESS will continue the galactic census — flagging more planetary candidates for further study.

Astronomers expect TESS to find about 20,000 planets in its first two years in operation, focusing on nearby, bright stars that will be easy for other telescopes to investigate later. About 500 of those expected exoplanets would be less than twice the size of Earth — and therefore may be good places to look for life.

The TESS mission is “a whole new opening for exoplanet studies,” MIT astronomer Sara Seager, TESS’ deputy science director, said during a news conference describing the upcoming launch.

TESS will be the first NASA science mission launched on the SpaceX Falcon 9 rocket. Once in orbit, the spacecraft will trace an unusual, elliptical path between Earth and the moon that will enable it to observe at least 85 percent of the sky — 350 times as much sky as Kepler saw.

Most of the planets found by Kepler orbit stars 1,000 light-years away or farther. TESS will focus on 200,000 stars that are a few hundred light-years away at most, and shine between 30 to 100 times brighter on average than Kepler’s.

The brighter the star, the easier it is to determine its planet’s characteristics, such as its mass and whether it has an atmosphere, Seager says. “Photons are our currency — the more, the better,” she says.

That follow-up will help TESS avoid some of Kepler’s pitfalls. Because Kepler’s stars were so far and so dim, some of its planet candidates were confirmed as actual planets only by statistics rather than by other telescopes. And not all those confirmations may stick. A recent paper posted at arXiv.org showed that Kepler 452b, an Earth-sized planet that orbits a sunlike star at the same distance Earth orbits the sun, may be a mirage (SN: 8/22/15, p. 16). Many of TESS’ planets won’t face the same uncertainty.

COASTING IN SPACE The TESS satellite’s unusual 13.7-day orbit uses the moon’s gravity to stabilize it, so it needs little fuel. During the part of the orbit colored blue, TESS will observe the sky. During the part marked in orange, it will transmit data back to Earth. The gray ring marks the moon’s orbit.

But the way TESS will search for exoplanets is the same as Kepler: The satellite will watch stars for signs of dimming, which can indicate that a planet is transiting, or crossing in front of, the star. Measuring how much starlight is blocked can tell astronomers the size of the planet.

Once TESS finds a planet, astronomers will need more information to understand its qualities, such as whether it’s rocky or gassy (SN Online: 6/19/17). For that, other telescopes will follow up. Ground-based telescopes will measure the gravitational tug of a planet on its host star to learn the planet’s density, which is a clue to its composition. Astronomers plan to measure masses for at least 50 TESS planets that are smaller than Neptune in the hopes that many of them will have rocky, and therefore potentially habitable, surfaces.

Before TESS, most known planets were more than 1,000 light-years away, with a few closer than 30 light-years (a parsec is 3.26 light-years). TESS (orange circles) will fill in the gap. The size of the circles represents how easy the planets are to find.

NASA’s James Webb Space Telescope, now scheduled to launch in 2020, will then check some of those planets for signs of life (SN: 4/30/16, p. 32).

“This is one of the major questions that TESS is intended to answer: Where will we be pointing Webb?” said the mission’s principal investigator, MIT astronomer George Ricker, at the press conference. Webb will peer at the starlight filtering through planetary atmospheres to try to detect molecules that could be produced by something living on the surface.

It will take a few months for TESS to swing into its regular orbit before it begins collecting data. At that point, it will be able to use the moon’s gravity to stabilize itself for decades in orbit without using extra fuel. The mission is set to last two years, but could continue taking data almost indefinitely.

“TESS is not going to be limited by any expendable or other aspects,” Ricker said. “It will be basically limited by how long NASA has the patience to fund the mission.”

NEW DELHI: Prime Minister Narendra Moditoday congratulated ISRO scientists for the successful launch of navigation satellite IRNSS-1I and said it will benefit the common man of the country.

ISRO’s navigation satellite INRSS-1I was today launched by PSLV-C41 from the spaceport at Sriharikota in Andhra Pradesh and was successfully placed in the designated orbit.

“Congratulations to our scientists on the successful launch of navigation satellite IRNSS-1I by PSLV. This success will bring benefits of our space programme to the common man. Proud of team @isro!” the prime minister tweeted.

The PSLV-C41/IRNSS-1I Mission blasted off at 4.04 am from the first launchpad at the Sathish Dhawan Space Centre. It was a normal lift-off, ISRO officials said.

Indian Space Research Organisation (ISRO)Chairman K Sivan described the mission as a success and congratulated the scientists.

He said IRNSS-1I was successfully placed in the designated orbit.

IRNSS-1I is expected to replace IRNSS-1A, the first of the seven navigation satellites, which was rendered ineffective after its three rubidium atomic clocks failed. The seven satellites are part of the NavIC navigation satellite constellation.

The launch is ISRO’s second attempt at sending a replacement satellite.
